Das Naturkundemuseum Karlsruhe zählt Dank seiner herausragenden Sammlungen und Forschungsaktivitäten zu den großen naturkundlichen Museen Deutschlands. Dazu trägt auch die zweifache Bedeutung des Hauses als wissenschaftliches Forschungsinstitut sowie als besucherorientiertes Museum bei. Auf über 4000 m2 Ausstellungsfläche werden verschiedene Bereiche der Naturwissenschaft vorgestellt, darunter einheimische und exotische Tiere aus verschiedenen Regionen der Erde in naturgetreu nachgebildeten Lebensräumen. Lebende Tiere aus dem Vivarium sind dabei als Ergänzung in die Ausstellungen integriert. Die Wissenschaftler aus den Abteilungen Botanik, Entomologie, Geologie/Paläonthologie und Zoologie sind weltweit in internationalen Forschungsprojekten engagiert – so ist es möglich, neben den Schausammlungen und Sonderausstellungen auch immer wieder die neuesten Ergebnisse aus der aktuellen Forschung zu präsentieren. Dabei spielt die Arbeit der Wissenschaftler nicht nur in der internationalen Forschung eine wichtige Rolle, sie leistet auch einen unmittelbaren Beitrag zum Arten- und Naturschutz. Die Veröffentlichungen und die Fachkompetenz der Wissenschaftler dienen der Arbeit von Naturschutzbehörden im In- und Ausland und tragen dazu bei, politische Entscheidungen in ökologischen und ökonomischen Fragen sachlich zu unterstützen. Die Insektensammlung des Naturkundemuseums ist sehr umfangreich und stellt mit ca. 2,5 Millionen Exemplaren die größte Einzelsammlung da. The Staatliches Museum für Naturkunde was founded 1791 and covers zoology, botany and palaeontology. Original, the collection base on the ducal collection of Baden-Wuerttemberg, which dates back to the 16th century. Over the centuries the collections grew and it still due. It comprises many different objects (e. g. more than four million fossils and insects) and is consult by several international specialists. Since 1985 the museum consist of two locations, the Museum am Löwentor and the Schloss Rosenstein. Most of the palaeontological collection is host in the Museum am Löwentor, furthermore most scientific collections are stored here. The Schloss Rosentein serves as exhibition area for aspects of the natural history. Four divisions (botany, entomology, palaeontology and zoology) conduct research on those four fields. The Stanford Human OligoGenome Project hosts a database of capture oligonucleotides for conducting high-throughput targeted resequencing of the human genome. This set of capture oligonucleotides covers over 92% of the human genome for build 37 / hg19 and over 99% of the coding regions defined by the Consensus Coding Sequence (CCDS). The capture reaction uses a highly multiplexed approach for selectively circularizing and capturing multiple genomic regions using the in-solution method developed in Natsoulis et al, PLoS One 2011. Combined pools of capture oligonucleotides selectively circularize the genomic DNA target, followed by specific PCR amplification of regions of interest using a universal primer pair common to all of the capture oligonucleotides. Unlike multiplexed PCR methods, selective genomic circularization is capable of efficiently amplifying hundreds of genomic regions simultaneously in multiplex without requiring extensive PCR optimization or producing unwanted side reaction products. Benefits of the selective genomic circularization method are the relative robustness of the technique and low costs of synthesizing standard capture oligonucleotide for selecting genomic targets. ...
